Have a 2008 Santa Fe with a 3.3 L V6. Check engine light started flashing,engine started running rough. Local parts Store said code showed number one cylinder misfire.

Put in some Lucas Oil, or clean boost max... Works for me.

Check the #1 spark plug , coil and fuel injector. Also do not drive it with the check engine light flashing as the raw fuel from the #1 cylinder can burn in the catalyst and melt the core of the catalyst. How many miles on this vehicle since the spark plugs were last changed? Also do not use a fuel additive on this problem expecting it to work.

Yes, #1 is at the left rear at firewall. How many miles on the engine, and have you ever changed the wires or plugs or had them off for any reason? As Bob says, check the wire and the plug and the coil first. It could be a defective or failing sensor as well, but most likely is a bad wire. Follow the wire from the #1 plug up to the coil to see which cone is the right coil, and check that both ends are firmly seated. If you have a dark enough place to put the car you may be able to run the engine in the dark and see if the wire or boots are arching out someplace along the length of it or at either boot. Let us know what you find.
